Tumor-infiltrating lymphocytes and other T cell-mediated treatments, such as CAR-T, have the potential to improve the effectiveness and personalization of cancer care. Cytokines play a crucial part in T cell survival, development, and differentiation.

T cell therapies typically entail the removal of autologous T cells from patients, cell manipulation/engineering, culture expansion, and re-infusion into the patient. A single administration of adoptive T cell therapy can necessitate billions of expanded T cells.

To produce the highest output of T cells while maintaining the correct phenotypic and function, high-quality cytokines must be used.

A powerful cytokine called interleukin-2 (IL-2) is frequently utilized in T cell culture to encourage the proliferation and differentiation of T cells into effector and memory cells.

Other c-cytokines, including IL4, IL-7IL-15, and IL-21, play a vital role in memory T cell development, proliferation, and survival. Varying combinations of IL-2, IL-7, IL-15, and IL-21 have been shown to efficiently expand T cells, resulting in populations expressing early-differentiation markers, such as CD27, CD28, CD62L, and CD127. 

To enhance a particular T cell phenotype, culture conditions can be adjusted. For instance, Th1 needs IL-12, Th2 needs IL-4, Tregs need TGF-β1, and Th17 needs IL-6.
