IFN-γ: A therapeutic target for cancer therapy

Interferon gamma (IFN-γ) is a cytokine that induces a series of immune responses against viruses and tumors. The pro-apoptotic, anti-tumor and anti-proliferative actions of the cells are activated when IFN-γ binds to the IFN-γ receptor, which is made up of IFNGR1 and IFNGR2.

IFN-γ expression that is erroneous encourages the growth of tumors. IFN-γ has thus become recognized as a major contributor to the immunogenicity of malignancy.

The CD4+ T helper cells and CD8+ cytotoxic T cells are the effector T cells that release IFN-γ. Natural killer cells, natural killer T cells, and B cells are additional immune cells that secrete IFN-γ.

IFN-γ is stimulated by interleukins such IL-12, IL-15, IL-18, and IL-21. These interleukins’ stimulatory effects also result from intracellular signaling of transcription factors such as AP-1, JAK/STAT, and T-bet.

In PD-1/PD-L1 immune checkpoint inhibitor cancer treatment, IFN-γ is a prognostic marker. The production of IFN-γ is associated with an improved response to immune checkpoint blockade (ICB) therapy in triple-negative breast cancer, and is considered to be an emerging high-potential adjuvant therapy target.
