The common gamma-chain cytokines communicate with the common cytokine receptor chain (γc/CD132/IL2RG) via cytokine receptors. At least six short-chain cytokines belong to this family: IL2, IL4, IL7, IL9, IL15 and IL21.
IL2RB (CD122), IL4R (CD124), IL7R (CD127), IL9R (CD129) and IL21R, along with IL2RG, are all members of the γc family of cytokine receptors. The γc subunit forms distinct heterodimeric receptors for IL4, IL7, IL9 and IL21, as well as heterotrimeric receptors for IL2 and IL15, by binding with these separate cytokine-specific receptor subunits.
This cytokine family regulates the formation, longevity, proliferation, specialization and function of cells both in innate and adaptive immune systems by activating three key signaling pathways (PI3K-Akt, RAS-MAPK and JAK-STAT).
