VEGFs and VEGF receptors: Key mediators in body systems

Vascular endothelial growth factors (VEGFs) are a family of secreted polypeptides that show angiogenic and mitogenic activities. The significance of VEGFs and receptors in tumorigenesis, angiogenesis, and neuroprotection makes them interesting targets for drug development.

The VEGF family members VEGF-A, VEGF-B, VEGF-C, VEGF-D, VEGF-E, VEGF-F, and PlGF attach in distinct patterns to three related receptor protein tyrosine kinases —VEGFR1/FLT1, VEGFR2/KDR, and VEGFR3/FLT4 — and two non-tyrosine kinase transmembrane proteins, neuropilin-1 (NRP1) and neuropilin-2 (NRP2).

The ligand-receptor attachment triggers the formation of homo- and heteromeric receptor complexes and enables downstream signaling.

Along with VEGF signaling, neuropilins also serve as co-receptors to various other signaling pathways, like those activated by Fibroblast Growth Factor (FGF), Insulin-like Growth Factor (IGF), Transforming Growth Factor beta (TGFβ), Hepatocyte Growth Factor (HGF), and Platelet-Derived Growth Factor (PDGF).

VEGF family ligands and receptors

More than 65 recombinant VEGF and receptor proteins that cover all VEGF family members and various isoforms of VEGF are provided by Sino Biological.
